The classic choices for imaging biological samples have been: low voltage field emission scanning electron microscopy, that can produce topographic images of samples with nanometer scale resolution, transmission electron microscopy providing potential sub-nanometer resolution of thin tissue cross-sections, and atomic force microscopy with sub-nanometer topographic resolution in all three dimensions, but with a limited depth of field.
Helium ion microscopy was carried out on an Orion helium ion microscope at 35 keV beam energy, with a probe current ranging from 0.1 to 1.5 pA. No conductive coatings were applied to the samples prior to imaging, in order to preserve the sample surface information.
The scanning of the helium ion beam and the formation of the image are synchronized so that for any given coordinate there is a corresponding signal or grey value in the resulting image.
In order to establish that the quality of these high magnification images is due to the HIM technology and not to the tissue processing method we employed, we also imaged these tissues by conventional scanning electron microscopy.

These challenges have recently been overcome with the development of the Helium Ion Microscope, which boasts advances in charge reduction, minimized sample damage, high surface contrast without the need for metal coating, increased depth of field and 5 angstrom imaging resolution.
One of the most formidable developments in imaging biological surfaces in their most native form without any chemical fixation is Cryo-SEM15, which freezes or cryo-immobilizes the sample and transfers it to a chilled stage prior to imaging.
To date, helium ion microscopy has been primarily been used in high-contrast imaging of inorganic materials; however23,24, there have been some limited instances where it has been used in imaging of biological samples.
As a model plant, Arabidopsis was chosen as a stable and well-studied organism to compare the effects of imaging at ever increasing magnification ranges between the FE-SEM and HIM. Since the samples were critical point dried and uncoated, they were prone to charging and thus low voltage imaging in the FE-SEM was required.
The critical point dried samples show especially clean surfaces, whereas the surfaces of the minerals in plunge-frozen and freeze-dried samples appear to have a much smoother envelope in the case of the uncoated samples.

The appellant is aggrieved by the judgment dated 30th April, 2019 passed by the High Court of Himachal Pradesh holding the appellant guilty of offence under Section 138 of the Negotiable Instruments Act, 1881 after reversal of the finding of acquittal returned by the learned trial Judge by its judgment dated 28th September, 2012.
On the basis of order placed by the appellant, nonwoven fabric was sold to the appellant vide invoice No.120 dated 01st October, 2010 and invoice No.135 dated 16th October, 2010 amounting to Rs.5,07,062/and Rs.5,10,000/which was delivered through public carrier truck bearing Nos.
On perusal of the evidence on record, the learned trial Judge returned a finding that the complainant failed to establish that the material/goods were delivered to the appellant in lieu of which, the cheques were issued, and in the absence of burden being discharged by the complainant, the onus to disprove or rebut the presumption could not be shifted to the appellant as referred under Section 139 of the Act.
Learned counsel for the appellant submitted that the complainant was not able to prove that the material/goods were ever sent or received by the appellant and in terms of the complaint, the burden was on the complainant to prove that the material/goods were received by the appellant, against which the cheques were received as security and even though the appellant has not placed any evidence to disprove or rebut the presumption in defence, still the complainant has to discharge its burden and has to stand on his own legs.
Learned counsel for the appellant further submits that the appellant was able to succeed in creating a doubt in the mind of the court below with regard to the nonexistence of the debt or liability, and the learned trial court had returned the finding based on the material available on record.
Once the facts came on record remained unrebutted and supported with the evidence on record with no substantive evidence of defence of the appellant to explain the incriminating circumstances appearing in the complaint against her, no error has been committed by the High Court in the impugned judgment, and the appellant has been rightly convicted for the offence punishable under Section 138 of the Act and needs no interference of this Court.
Thereafter, two separate legal notices were served by the complainant which were duly received by the appellant and even after receiving the said notices, the appellant neither responded to the notices nor made any payment within the statutory period of fifteen days and only thereafter, two separate complaints were filed by the complainant under Section 138 of the Act against the appellantaccused.

Subsequently, the complainant approached the High Court which affirmed that the primary burden was discharged by complainant that the cheques were issued by the appellant for the materials supplied.
Where any cheque drawn by a person on an account maintained by him with a banker for payment of any amount of money to another person from out of that account for the discharge, in whole or in part, of any debt or other liability, is returned by the bank unpaid, either because of the amount of money standing to the credit of that account is insufficient to honour the cheque or that it exceeds the amount arranged to be paid from that account by an agreement made with that bank, such person shall be deemed to have committed an offence and shall, without prejudice to any other provisions of this Act, be punished with imprisonment for , or with fine which may extend to twice the amount of the cheque, or with both: Provided that nothing contained in this section shall apply unless-.
The drawer of such cheque fails to make the payment of the said amount of money to the payee or, as the case may be, to the holder in due course of the cheque, within fifteen days of the receipt of the said notice.
Section 139- Presumption in favour of holder:-It shall be presumed, unless the contrary is proved, that the holder of a cheque received the cheque of the nature referred to in section 138 for the discharge, in whole or in part, of any debt or other liability.
v. Magnum Aviation Pvt. Ltd. contended that there is absence of prima facie burden being discharged by the complainant and mere issuance of cheque would not be sufficient to justify that the cheques were issued in discharge of the debt.
The drawer of the cheque will be held liable to prosecution on dishonour of cheque.
CONCLUSION. As observed by the court in this judgement itself the object of the provision laid down in section 138 of NI Act is to enhance the acceptability of cheques in the settlement of liabilities and in case where the person issuing the cheques does not exhibit the intention to honour the cheque, he shall be liable on dishonour of the same.

Because many government organizations require physical copies of your fingerprints, many IdentoGO Centers offer the ability to digitally collect an applicant’s fingerprint images and then print them onto a standard fingerprint card.
Ordering Fingerprint Cards And Training Aids — FBI
In order to request fingerprint cards and training aids, you must have an Originating Agency Identification Number.
Once you have been assigned an ORI Number, complete the 1-178, CJIS Supply Requisition Form, to order fingerprint supplies and/or the 1-178a, CJIS Fingerprint Training Aids Supply Form, to order training materials.
Agencies must identify their ORI Number on the requisition forms in order to receive fingerprint supplies and training aids.
Can I receive fingerprint cards without the ORI Number pre-printed on them?
Yes, agencies may request fingerprint cards with or without their ORI Number pre-printed on them.
One column is for the quantity without the ORI Number and one column is for the quantity to be pre-printed with the ORI Number.
There is no charge for the fingerprint cards and training aids.

Arizona Department Of Public Safety
Applicants can now utilize the Public Service Portal to apply for a Fingerprint Clearance Card.
The new secure PSP will allow applicants the ability to fill out and submit an FCC application online and pay the associated fees directly to the Arizona Department of Public Safety via a secure credit card checkout.
Electronic fingerprinting is only available if the applicant is physically present in Arizona.
If the applicant selects to submit prints via a paper fingerprint card, they will be sent a fingerprint card in the mail for use, or they can use a card supplied by a fingerprint vendor.
Once AZDPS receives the print cards or electronic prints, the fingerprints will be processed for review.
Applicants applying for Identity Verified Prints cards, and who select to submit their fingerpritns via paper, will be sent the blue postage-paid return envelope with the fingerprint cards.
Applicants renewing a valid IVP will be able to do so entirely on the PSP. Applicants will existing cards will be able to create an account and link their existing cards to their accounts.
