For most of the past 25 years, Jeff Sagansky has followed the same ritual every morning.
In a stellar career that has also included stints running several movie studios and a national theater chain, Sagansky has always had his heart in television.
Of his boyhood in Wellesley, Massachusetts, in the 1950s, Sagansky recalls that TV characters like the Cartwrights in Bonanza “Became an important part of my life.” In high school, he followed the industry in Variety.
Armed with his MBA, Sagansky spent a year as an analyst at CBS in New York before heading to the West Coast, where he worked at NBC and then with an independent production company.

New Delhi: The Income Tax Department has found that Hero MotoCorp made more than Rs 1000 crore bogus expenses and over Rs 100 crore cash transactions for a farmhouse in Chhattarpur, Delhi, sources told ANI. Income Tax Department carried out a search and seizure operation on March 23 on Hero Motocorp and its chairman and managing director Pawan Munjal at multiple locations in Delhi NCR which concluded on March 26.The search operations covered more than 40 premises spread over different locations in Delhi NCR. Sources told ANI that a large number of incriminating evidence in the form of hard copy documents and digital data have been seized during the search operations.
These evidences revealed that the group has booked bogus purchases, made huge unaccounted cash expenditures and obtained accommodation entries, aggregating to the tune of more than Rs 1000 crore.
The Department has also found evidence of cash transactions of more than Rs 100 crore in the purchase of a farm house in the outskirts of Delhi.
Munjal purchased a farmhouse in Chhattarpur where the market price of farm house was manipulated to save tax and used black money to pay cash more than Rs 100 crore which is a violation of section 269 SS of IT Act.
According to section 269SS of Income Tax Act, while transacting Immovable Property, 100 per cent penalty will be levied if the seller has accepted an amount of Rs 20,000 or more in cash from the buyer.
Last week raids were being conducted at the offices and residential premises of promoters of Hero MotoCorp. The search includes the residential and official premises of Pawan Munjal.
Statements of those present at the place where searches are being conducted were recorded and panchnama was done.

The Income Tax Department said two-wheeler manufacturer Hero MotoCorp made over Rs Rs 1,000 crore in bogus expenses and found multiple violations amounting to Rs 100 crore in transactions conducted for a farmhouse in Delhi.
The development comes after the department on March 23 conducted search and seizure raids at locations in Delhi-NCR on the premises related to Hero MotoCorp and its managing director Pawan Munjal and other top officials of the company.
According to news agency ANI, the I-T Department seized incriminating evidence in the form of hard copy documents and digital data during the raids conducted earlier.
The evidence collected revealed the two-wheeler manufacturer booked bogus purchases, made huge unaccounted cash expenditures and obtained accommodation entries amounting to Rs 1,000 crore.
The ANI report further added that I-T Department also found evidence of cash transactions of more than Rs 100 crore in the purchase of a farmhouse on the outskirts of Delhi.
Munjal purchased a farmhouse in Chhattarpur where the market price was manipulated to save tax and used black money to pay Rs 100 crore in cash in violation of section 269 SS of the IT Act, it added.

Betances, 34, can earn a prorated portion of a $2.75 million base salary if he reaches the major leagues, with an additional $500,000 in performance bonuses, according to the New York Post.
Betances was one of the sport’s most dominant relievers while with the New York Yankees from 2014 to 2018, posting a 2.22 ERA with 607 strikeouts in 373⅓ innings.
Betances has been off a mound this offseason, but the Dodgers need a longer look to determine how far he is from tapping back into some of his prime form.

All that is required on the plasmid DNA is an origin of replication-the DNA sequence that signals the host cell DNA polymerase to replicate the DNA molecule.
The most common method of DNA sequencing is based on premature termination of DNA synthesis resulting from the inclusion of chain-terminating dideoxynucleotides in DNA polymerase reactions.
Essentially, DNA polymerase is used for repeated replication of a defined segment of DNA. The number of DNA molecules increases exponentially, doubling with each round of replication, so a substantial quantity of DNA can be obtained from a small number of initial template copies.
The starting material can be either a cloned DNA fragment or a mixture of DNA molecules-for example, total DNA from human cells.

Biological Principles
Molecular biologists coined the term “Molecular cloning” to describe the process of selectively replicating a chosen segment of DNA. The cloned DNA segment may be replicated within a cell, using “Recombinant DNA” technology, or in a test tube, using the polymerase chain reaction.
Even random DNA segments, where the base sequences are unknown, may be amplified by ligating adapter primers, short synthetic DNA segments of known sequence, to the ends of the target DNA molecules.
The last feature is important because the ligation of plasmid and foreign DNA segments favors the plasmid ends re-ligating without a foreign DNA insert, resulting in the original, “Empty” plasmid with no foreign DNA insert.
Cloning into the 5′ end of the lacZ gene also means that the E. coli cell can express a protein encoded by the inserted DNA. The lac promoter provides a means to regulate transcription, and protein coding sequences in the inserted DNA can be expressed as a fusion protein, containing the first few amino acids of the E. coli beta-galactosidase gene, and any amino acids encoded in the same reading frame by the inserted DNA sequence.
Cas9 targets the DNA site to be cut by using a short guide RNA. Cas9 binds the sgRNA, and cuts DNA wherever the sgRNA binds to a complementary DNA sequence.
After Cas9 cuts the DNA to create a double-strand DNA break, the cell’s DNA repair system will trim the broken ends and ligate them together, often creating small deletions as a result of the trimming.
If a homologous DNA sequence is available, the cell’s DNA repair system uses the matching DNA as a template to repair the break in the DNA. This homology-dependent repair system copies the sequence information in the DNA template as it joins the broken ends together.
Recombinant DNA
Plant DNA may be joined to bacterial DNA, or human DNA may be joined with fungal DNA. In addition, DNA sequences that do not occur anywhere in nature may be created by the chemical synthesis of DNA, and incorporated into recombinant molecules.
Using recombinant DNA technology and synthetic DNA, literally any DNA sequence may be created and introduced into any of a very wide range of living organisms.
Vectors are generally derived from plasmids or viruses, and represent relatively small segments of DNA that contain necessary genetic signals for replication, as well as additional elements for convenience in inserting foreign DNA, identifying cells that contain recombinant DNA, and, where appropriate, expressing the foreign DNA. The choice of vector for molecular cloning depends on the choice of host organism, the size of the DNA to be cloned, and whether and how the foreign DNA is to be expressed.
In standard cloning protocols, the cloning of any DNA fragment essentially involves seven steps: Choice of host organism and cloning vector, Preparation of vector DNA, Preparation of DNA to be cloned, Creation of recombinant DNA, Introduction of recombinant DNA into the host organism, Selection of organisms containing recombinant DNA, and Screening for clones with desired DNA inserts and biological properties.
This can happen, for example, when a recombinant DNA fragment containing an active promoter becomes located next to a previously silent host cell gene, or when a host cell gene that functions to restrain gene expression undergoes insertional inactivation by recombinant DNA. Recombinant DNA is widely used in biotechnology, medicine and research.
The first publications describing the successful production and intracellular replication of recombinant DNA appeared in 1972 and 1973, from Stanford and UCSF. In 1980 Paul Berg, a professor in the Biochemistry Department at Stanford and an author on one of the first papers was awarded the Nobel Prize in Chemistry for his work on nucleic acids “With particular regard to recombinant DNA”.
Scientists associated with the initial development of recombinant DNA methods recognized that the potential existed for organisms containing recombinant DNA to have undesirable or dangerous properties.
