The Toyota Supra is a sports car and grand tourer manufactured by the Toyota Motor Corporation beginning in 1978.

Ever since, the rumor mill has long speculated a stick-shift Supra was in the works, and today, Toyota’s European division finally confirmed that the 2023 Toyota Supra will be getting a “All-new, tailor-made” manual transmission model.

The Supra is currently on sale with either a 255-horsepower turbocharged 2.0-liter I-4, or a 382-horsepower turbocharged 3.0-liter I-6, and the rumor mill suggests the new transmission will likely only be equipped on the latter, more powerful model.

Judging from previous exec comments, it’s also possible that the manual GR Supra could just be a limited run, as future special editions of the car have been hinted at before.

Toyota says the “Toyota GR Supra Manual Transmission has been engineered to offer enthusiasts something closer to the GAZOO Racing DNA that also enhances the driving experience.” The tease suggest more information will be available in a few weeks time, finishing off with a “Save the Manuals” hashtag.

Toyota USA also teased the new manual Supra on Twitter, so it’s likely coming stateside, as well.

Square Enix’S Latest RPG Falls Below 10 Players On Steam

Babylon’s Fall, the latest co-op action RPG from Square Enix and Platinum Games, struggled to attract an audience when it launched last March, and has fallen below 10 concurrent players on PC for the first time.

As game-tracking site SteamCharts shows, only eight players booted up Babylon’s Fall on Steam during one point on April 13, marking a new low for the ailing game.

Players have been drifting from the game since it launched at the beginning of March.

“We will continue to provide new content for the game and make improvements based on player feedback, striving to keep existing players playing and to attract newcomers.”

Square Enix will likely be disappointed with the sales of Babylon’s Fall, but its meager player count also undermines the game’s live-service business model.

Without a large player count, it will be a struggle to get a team together to play through the additional content.

Dice’s multiplayer shooter has now fallen below 1,000 concurrent players on Steam, with many moving to an indie Battlefield clone for their squad-based FPS fix.

Xbox Game Pass Gets New Award-Winning Square Enix Game

Xbox Game Pass has added another notable title to the ever-expanding subscription platform, this time in the form of an award-winning game from publisher Square Enix.

Over the past month, Square Enix has been bringing a handful of different titles to Xbox Game Pass, notably with the addition of Marvel’s Guardians of the Galaxy.

As of this moment, Life Is Strange: True Colors is available to download for Xbox Game Pass subscribers.

The latest entry in the long-running narrative-driven series has come to both Game Pass verticals, which means that those on Xbox One, Xbox Series X, Xbox Series S, and PC can look to play it for themselves.

“Alex Chen has long suppressed her ‘curse’: the supernatural ability to experience, absorb and manipulate the strong emotions of others, which she sees as blazing, colored auras,” says the official description of Life Is Strange: True Colors from Square Enix.

Some of those accolades include the award for Best Storytelling at the Golden Joystick Awards and the Games for Impact honor at The Game Awards.

Kingdom Hearts IV Announced By Square Enix

Square Enix announced Kingdom Hearts IV today during a 20th anniversary event for the franchise.

That entry came out over a decade after the release of 2005’s Kingdom Hearts II. However, Square Enix released multiple spin-off titles between those numbered entries.

We’ve only had one spin-off, the 2020 music game Kingdom Hearts: Melody of Memory, release after Kingdom Hearts III so far.

Square Enix also revealed Kingdom Hearts: Missing-Link at the event, a more modest title that appears to be a prequel story.

The realistic city setting is not a surprise to any Kingdom Hearts fans who watched KH3’s secret ending or played the game’s DLC story, Re Mind.

Square Enix noted before that KH3 was the last chapter of the series’ first story arc, which centered around the villain Xehanort.

KH4 is continuing plot threads introduced in the mobile/browser game Kingdom Hearts χ, while picking up from KH3’s cliffhanger ending.

A modern core drill is a drill specifically designed to remove a cylinder of material, much like a hole saw.

In 1983, the first successful clinical trial utilized an anti-carcinoembryonic antigen antibody-vindesine conjugate []. Patient specific antibody drug conjugates increased therapeutic value by decreasing chemotoxicity, restricting the systemic circulation of destructive agents without restricting their activity on the tumor tissue.

The unique property of antibody-drug conjugates is that these so-called armed antibodies selectively dispatch highly potent cytotoxic anticancer chemotherapies directly to cancer cells while, at the same time, leaving healthy tissue unaffected.

Antibody-Drug Conjugates deliver highly potent cytotoxic anticancer agents to cancer cells by joining them to monoclonal antibodies by biodegradable, stable linkers and discriminate between cancer and normal tissue.

Advances in linker technology needed to attach monoclonal antibodies to cytotoxic anticancer agents to allow control over drug pharmacokinetics and significantly improve delivery of a cytotoxic agent to cancer cells.

In the lysosome, proteases in a mild acidic environment digest the monoclonal antibody to release free payloads, which then cross the lysosome membrane to enter the cytoplasm and/or the nucleus where they bind to the target molecule which, in turn, leads to cell death.

The cytotoxicity of the inhibitors of tubulin polymerization such as the maytansinoids, dolastatins, auristatin drug analogs, and cryptophycin are related to their ability to inhibit cell division by binding tubulin, which arrests the target cell in the G2/M stage of the cell cycle resulting in apoptosis.

Although the concept of an ADC seems simple, designing a successful ADC is complex and requires careful selection of the receptor antigen, antibody, linker, and payload. In this review, we explore insights in the antibody and antigen requirements needed for optimal payload delivery and support the development of novel and improved ADCs for the treatment of cancer and neurological diseases.

Choosing the right target antigen and antibody combination can play an equally important role in ADC development.

The target antigen, necessarily present on the cell surface, should be internalizing to enable the ADC, following binding, to be efficiently transported into the cell through receptor-mediated endocytosis prior to active drug release in, for example, the lysosome.

The linkage between the antibody and the cytotoxic payload is a critical component of an ADC and requires careful design and development.

Mechanistically, an ADC acts by binding to the target antigen on the cell surface, followed by its internalization via antigen-mediated endocytosis, trafficking into the lysosome, and the release of the payload through the proteolytic degradation of the antibody moiety and/or cleavage of the linker.

1 The rationale for developing an ADC is that linking a cytotoxic agent to a tumor-targeting antibody will enable its selective targeting to cancer cells, leading to their eradication while sparing cells in normal tissues.

ADC specificity is thus provided by the targeting antibody rather than by blocking key signal transduction nodes uniquely required for tumor growth or survival.
